Overview
A long-acting synthetic GnRH agonist whose extended half-life produces sustained GnRH-receptor signaling that paradoxically suppresses LH and FSH — the 'pulse paradox' applied therapeutically. Used clinically to suppress gonadal hormone production in conditions like prostate cancer, central precocious puberty, and endometriosis.
Mechanism of action
Triptorelin is a modified GnRH analog with a longer half-life than native gonadorelin. Continuous receptor activation first causes a transient surge of LH/FSH, then downregulates and desensitizes pituitary GnRH receptors, leading to sustained suppression of gonadotropin release and consequently a sharp drop in testosterone (or estrogen). This is the same pulse-paradox mechanism as native GnRH, engineered into a practical long-acting therapeutic for chemical castration and hormone suppression.
