Acetylation and Amidation: Protecting a Fragile Tetrapeptide
Epitalon's AEDG sequence is only four amino acids — which makes it fast-acting but also fast-degraded. Exopeptidases clip peptides from their ends, and a four-residue peptide has almost no interior to protect. N-acetyl Epitalon Amidate solves this by capping both termini: the N-acetyl group blocks amino-exopeptidases, the C-terminal amide blocks carboxy-exopeptidases.
It's the same engineering principle as PEG-MGF or mod GRF(1-29): when a peptide's mechanism is good but its half-life is the problem, modify the molecule rather than abandon it. The underlying telomerase biology — and its theoretical cancer consideration — is unchanged from epitalon. Members discuss the modified variant mostly in terms of pharmacokinetic practicality.
