Overview
Native gonadotropin-releasing hormone (GnRH) that stimulates the pituitary to release luteinizing hormone (LH) and follicle-stimulating hormone (FSH), central to reproductive hormone research and fertility testing. Like several native peptide hormones, it requires pulsatile delivery — continuous administration paradoxically suppresses the very hormones it stimulates.
Mechanism of action
Gonadorelin binds GnRH receptors on pituitary gonadotrophs, triggering pulsatile release of LH and FSH — the gonadotropins that drive testosterone production in the testes and estrogen/ovulation in the ovaries. Crucially, this is pulse-dependent: the pituitary interprets a continuous GnRH signal as 'shut down' rather than 'stimulate,' which is why GnRH analogs with long half-lives are used to suppress rather than stimulate gonadal function. Native pulsatile GnRH preserves the physiological stimulation pattern.
