Overview
5-Amino-1MQ is a small-molecule (non-peptide) inhibitor of nicotinamide N-methyltransferase (NNMT), an enzyme upregulated in obesity and certain cancers. By inhibiting NNMT it is theorized to increase cellular NAD⁺ and shift metabolism toward energy expenditure and fat oxidation, studied in preclinical obesity models.
Mechanism of action
NNMT methylates nicotinamide, consuming the methyl donor SAMe and reducing NAD⁺ availability. 5-Amino-1MQ inhibits NNMT, which is proposed to preserve NAD⁺ and methyl-donor pools, increase SIRT1/NAD⁺-dependent metabolic activity, and shift adipocyte metabolism toward lipolysis and energy expenditure — reducing fat accumulation in preclinical models.
