Overview

Teriparatide is the recombinant N-terminal (1-34) fragment of human parathyroid hormone (PTH) and an FDA-approved anabolic agent for osteoporosis. Unlike antiresorptive drugs that slow bone loss, teriparatide actively stimulates osteoblasts to build new bone when administered intermittently, and is also studied for fracture healing.

Mechanism of action

Teriparatide mimics the N-terminal active fragment of parathyroid hormone. When given intermittently (rather than continuously), it stimulates osteoblast activity and net new bone formation, increasing bone mineral density and reducing fracture risk — an anabolic rather than antiresorptive mechanism.
