Most antidepressants work slowly — SSRIs take weeks to remodel serotonin signaling, and that lag is a clinical problem. PE-22-28 comes from a different hypothesis: what if you could change serotonergic neuron excitability directly, fast?

TREK-1 is a potassium channel that sets the resting potential of certain serotonergic neurons. When it's open, the neuron is harder to fire. PE-22-28 blocks it, making the neuron more excitable and boosting serotonergic transmission. Preclinical TREK-1-knockout animals are resistant to depression-like behavior, which is the genetic evidence supporting the target. The appeal is speed — a channel block is fast in a way weeks-long SSRI remodeling isn't. The catch: it's all preclinical so far.
