Ciliary neurotrophic factor (CNTF) is a powerful neurotrophic and neurogenic signal — but as a full cytokine it caused unacceptable weight loss and muscle effects in clinical trials, killing its development. P21 is the engineering answer: a small mimetic designed to reproduce CNTF's beneficial neurotrophic signaling through the gp130/JAK-STAT pathway while shedding the systemic effects.
