Overview
A 10-amino-acid fragment of kisspeptin that sits upstream of GnRH in the reproductive axis, acting on kisspeptin receptors (KISS1R/GPR54) on GnRH neurons to drive GnRH release and downstream LH/FSH. Discovered through the gene KISS1 (originally named for metastasis suppression), kisspeptin is now understood as a master gate of puberty and reproductive function.
Mechanism of action
Kisspeptin-10 binds the kisspeptin receptor (KISS1R, formerly GPR54) on GnRH-secreting hypothalamic neurons, stimulating GnRH release and thereby the entire downstream reproductive cascade (LH, FSH, gonadal steroids). The importance of this pathway was revealed by a natural experiment: humans and mice with KISS1R loss-of-function fail to undergo puberty, establishing kisspeptin signaling as the upstream gate that permits reproductive activation. KP-10 is the shortest active fragment with full receptor efficacy.
